Casey Jones joins the roster of playable characters after you roll the game's credits once, which entails you clearing all sixteen episodes and putting an end to the final boss. Once the credits stop scrolling, Casey will be revealed as an available character for you to play.
Even with the requirement of getting Casey Jones has you finish the game once, it's not as bad as it sounds. The average first playthrough of Teenage Mutant Ninja Turtles: Shredder's Revenge takes roughly two and a half hours to complete, which is doable in one sitting.
